ENHANCING LIGNIN PEROXIDASE PRODUCTION USING PURPUREOCILLIUM LILACINUM

Abstract

A fermentation method is selected for the laboratory scale production of lignin peroxidase enzyme Purpureocillium lilacinum by using student t-test. The P-value was found to be 0.001 which indicates solid state fermentation (SSF) method is better than submerged fermentation (SmF) method. The substrate for SSF fermentation is selected by using one way ANOVA. The calculated P-value corresponding between groups is very low (< 0.05) indicates the production of lignin peroxidase enzyme is not same for all substrates, by using OFAT Wheat straw containing media shows maximum activity 32.05U/ml. Optimization of wheat straw containing media was done using OFAT for physical parameter. Purpureocillium lilacinum (LP8i) maximum activity at the 11th day of incubation (182.39U/ml), pH 4.00 (53.46U/ml), temperature 20 0C (46.16U/ml), moisture content 60% (50.83 U/ml) was noted for these physical parameters.
