QUANTIFICATION OF PROTEIN AND -AMYLASE ACTIVITY IN FOUR DIFFERENT VARIETIES OF SORGHUM (SORGHUM BICOLOR)RASHMI KARANI, SHRISHAILNATH SAJJAN, SUHASINI CHIKKALAKI AND SUMA PATTARAbstract Protein quantification and -amylase activity (sorghum bicolor) was carried out in four different varieties of sorghum. Seeds of varities viz., DSV 4, DSV 5, CSV 216R and M35 were procured and washed with pinch of mercuric chloride and repeatedly washed with distilled water were sown in sterile soil medium in pots. Quantification of protein is done through Folin-Cio-Calteau method. DSV-5 showed the highest concentration at 660 nm and M-35 showed least concentration in standard BSA graph. The transparent bands were observed immediately against dark background. The transparent bands indicated the presence of amylase enzyme which degraded the starch molecules. Amylase is forms were prominent in shoot samples and faint bands were observed in root samples.
