GENETIC DETECTION AND SEQUENCING OF SOME VIBRIO TOXIN GENES ISOLATED FROM DIARRHEAL PATIENTS

Abstract

Detection of some virulence genes and sequencing of some genesin Vibrio cholerae. A total of 200 samples, only 25 V. cholera isolates were obtained from diarrheal samples which detect by typical bacteriological and serological approaches. Also, four virulence genes (ctxAB, hly, ace, and ompW) were investigated in 25 isolates by using polymerase chain reaction (PCR) and sequencing analyze was done by used ace gene. In the present study, it was refer that hly gene was found in all isolates understudy, Moreover, 15 isolates were show to possess ace gene in their genome. On the other hand, only nine isolates of V. cholera were show to possess ctxAB gene also it was found that 21 isolates contain ompW gene. sequencing of ace gene of V. cholerae was done and the results exhibited identity 94-100% homology to the sequences of standard strain Vibrio cholera strain 62013 accessory cholera enterotoxin ace gene. These findings reinforce the connection between virulence factors and human infection, nucleotide sequence founded some differentare present which trends to horizontal gene transmission among the V. cholera isolates have assumed rise to appearance of original pathogenic strains. V. cholera strains has a role played by ctxAB, ompW, hly, and ace in cholera contagion in humans.
