Abstract

This study aims to determine the sensitivity of Escherichia coli bacteria to antibiotics and to detect encoding gene tetA and sull using the Polymerase Chain Reaction technique. This project uses two antibiotics that will be tested for sensitivity by the diffuse disc method, namely tetracycline and sulfonamide. In this study,86 of 128 positive fresh milk samples contained Escherichia coli bacteria that had been planted in EMBA media and confirmed by the Indol test. The results of the electrophoresis of PCR products stated that E. coli, which was resistant to antibiotics was indicated by the presence of bands. The 12 positive samples contained Escherichia coli and resistant to tetracycline’s with 11 samples which showed positive band DNA, resulting from tetA primary amplification with 178 bp long. Furthermore, 12 samples of Escherichia coli bacteria that were resistant to sulfonamides were run by PCR, there were 5 positive samples of DNA bands from sull primers with a length of 160 bp.
