ANTIOXIDANTS, DNA DAMAGE PROTECTIVE PROPERTIES ANDANTIBACTERIAL ACTIVITY OF LEAF EXTRACT OF PHLOGACANTHUS THYRSIFLORUS NEES

Abstract

Methanolic leaf extract of Phlogacanthus thyrsiflorus Nees was investigated for total polyphenol, flavonoid and anthocyanin content (TPC, TFC and TAC respectively). In vitro antioxidant activities were measured using Ferric reducing antioxidant power (FRAP) assay, 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2-2'-Azino-bis-(3-ethylbenzo-thiazoline-6-sulfonic acid) diammonium salt (ABTS) radical scavenging capacity assay. DNA nicking assay and antioxidant activities of extract were carried out. Polyphenol content particularly flavonoids were found high in the extract. The extract exhibited significant antibacterial activity for the human pathogenic bacteria. The ability of extract to protect DNA damage caused by UV induced photolysis of H2O2 using pUC18 plasmid was found significant at concentration of 1mg mL-1 and equivalent to the standard antioxidant Quercetin (100μm). The high concentration of flavonoids in the leaf extract are likely to be responsible for high antioxidant activities as well as antibacterial activity in P. thyrsiflorus.
