Abstract
Broad genetic diversity of Adenium obesum is the key for genetic improvement of this important ornamental species.Use of seventeen arbitrary ten decamer oligonucleotide primers of four series viz. OPA, OPI, OPJ, OPN generated total 700 scorable bands with 155 loci, among them 148 loci were found polymorphic, showing 95% polymorphism. On an average, per primer 9 polymorphic loci were generated. The RAPD primers tested in present investigation produced fragments of different sizes. The minimum (121 Base pair) sized fragment was amplified by primer OPN-7, whereas maximum (2547 base pair) sized fragment was amplified by the same primer i.e. OPN-7. A highest (100%) polymorphism was exhibited by primers OPA- 1, OPA-9, OPJ-13, OPJ-17, OPI-19, OPI-20, OPJ-5, OPN-4, OPN-5, OPN-6, and OPN-7 while the lowest polymorphism percentage was recorded by two primer, i.e. OPA-18 and OPJ-121 with polymorphic value value of 66.66 per cent. Thus entire study revealed that genotype âDeang Udam Sapâ and âPicotteâ were found to be more genetically divergent genotypes followed by genotypes âDeang Udam Sapâ and âMor Lok Dokâ. Genotypes âDouble Sweet heartâ and âVithoonâs Whiteâ were found to be genetically closest genotypes flowed by genotypes âMor Lok Dokâ and âPicotteâ. Genetic similarity between genotype âArrogantâ and âMung Siam Purpleâ was also found to be considerable.
