ISOLATION OF LYSOBACTER SP. FROM SOIL AND OPTIMIZATION OF L-GLUTAMINASE PRODUCTION

Abstract

A bacterium Lysobacter sp. was isolated from local soil and evaluated for the production of Lglutaminase considering the potential application of the L-glutaminase in chemical, pharmaceutical and food industries. The screening and selection of the bacteria were carried out on the chitin agar medium by the appearance of pink color colonies. The soil isolate found to have the capability to produce L-glutaminase and was assayed by direct Nesslerization measured at 425nm. The enzyme was produced by submerged fermentation on different solid substrates such as wheat bran, rice bran, black gram bran, corn bran, pigeon pea bran and pea bran. Optimization of the enzyme production was carried out done with one factor at a time and Response Surface Methodology. The maximal enzyme production was observed using wheat bran as solid substrate and the activity was 0.9IU/mL.
