ASSESSMENT OF GENETIC DIVERSITY OF DIFFERENT GROUNDNUT (ARACHIS HYPOGAEA L.) GENOTYPES BY USING MOLECULAR MARKER

Abstract

The present study was carried out to assess the genetic diversity by using 10 simple sequence repeat (SSR) markers. DNA was extracted from tender young leaf samples collected from eight groundnut varieties. A protocol using 0.090 g mL-1 glucose, 0.010 g mL-1polyvinylpyrrolidone, 0.0040 g mL-1 sodium bisulphite, 0.0050 g mL-1 sodium dodecyl sulphate, and 50 μL mL-1sarcosine produced good quality DNA. A total of 88 scorable DNA fragments were produced and among them 80 DNA fragments were found to be polymorphic between eight varieties of groundnut. The minimum number (8) of polymorphic fragments produced by pPGPseq2F05, pPGPseq8E12, pPGSseq13A10, pPGSseq16C06, pPGPseq2B10, PM384, PM137, PM003, while the maximum number of polymorphic fragments were found to be 16 (pPGPSseq17F06) and 8 average number of polymorphic bands observed per primer. The average polymorphism percentages revealed using SSR markers between the eight varieties were 90%. Using SSR markers, the overall similarity between all five varieties ranged from 0.095 to 0.375. SSR marker analysis proved that SSR marker is a powerful tool for assessment of genetic diversity between groundnut varieties. Identification of genetically diverse genotypes will help in future breeding programs of groundnut.
