A STUDY ON ANTI-FUNGAL ACTIVITY OF KOJIC ACID SYNTHESIZED BY ASPERGILLUS FLAVUS FJ537130 STRAIN ISOLATED FROM PEANUT SOIL

Abstract

A natural organic anti-metabolite compound kojicacid had a good antimicrobial properties and used widely in cosmetic applications. In the present investigation this compound kojic acid was extracted by using various cheap carbon substrates using a novel strain Aspergillus flavus screened from soil. 18Sr RNA analysis using NCBI database was used for the strain identification. The highly purified compound graded by HPLC was tested on yeast and fungi isolated from patient samples with dermatophytic infections. Surface fermentation was performed with 14 different solid and liquid substrates by using Aspergillus flavus FJ537130. The final product was purified and crystallized. The crystalline powder of kojic acid was liquefied to a specific working volume concentration and used for the anti-fungal activity studies through Anti-fungal Disk diffusion method. Out of 50 clinical samples screened 8 samples showed positive growth. The fungi were isolated using standard microbiological methods from various patient samples suffering with dermatophytic infections. The organisms isolated were Trichophyton species, Candida albicans, Aspergillus fumigatus, Aspergillus terreus, Mucor, The compound showed maximum inhibitory activity against Candida albicansisolated from 70 years old patient.
