IDENTIFICATION OF HYBRID AND GENETIC PURITY ASSESSMENT IN COTTON (GOSSYPIUM SPP.) USING MICROSATELLITE MARKERS

Abstract

Genetic purity is one of the most important characteristics of quality seed. In order to distinguish the parents from hybrid and to identify impurities in parental lines the current investigation was carried out using Simple Sequence Repeat (SSR) markers at seed Research and Technology Centre, Rajendranagar Hyderabad. About 50 loci were selected to identify specific markers associated with genetic purity testing of hybrid Phule-399 and their corresponding parents (RHCB009, female parent) and (RHCB001, male parent) to replace grow out test. It was observed that JESPR 151 could produce complementary banding pattern between patents and hybrids by amplifying female line at 160 bp and male parent at 120bp. Further, a simple procedure has been standardized utilizing a two dimensional bulked DNA sample strategy involving a 20×20 grow out matrix which was designed for identification of contaminants (off types) in the seed lot of parents lines.
