COMPARATIVE STUDIES ON DEGRADATATION OF CELLULOSE IN COIR PITH USING THE FUNGUS PLEUROTUS SAJOR CAJU AND TRICHODERMA VIRIDE

Abstract

The cellulytic fungus Pleutrorus sajor caju and Trichoderma viride (parent and mutant) produced extracellular degrdative enzymes such as filter paper cellulase (FPCase) endoglucanse, cellobiase and Xylanase. The fungal strains were capable of degrading cellulosic content in coir pith. Among the fungus Trichodernui viride mutant that are able to produce higher level of cellulases. The nitrogen sources, potassium nitrate increase the activity of FPCase, endoglucanase, cellobiase and xylanase than the urea and ammonium sulphate. The reaction mixture was incubated at 50°C for 30 min. Dinitro salicylic acid (DNS) was used for measured the reducing sugar (pH 4.8) FPCase, endoglucanase and cellobiase activitites were maximum on 8th, 16th and 24th day of incuabtion respectively. The xylanase activity was maximum on 8'h day of incubation.
