UTILIZATION AND OPTIMIZATION. OF RENEWABLE RESOURCES FOR THE STABLE PRODUCTION OF CELLULOSE DEGRADING FUNGAL ENZYMES

Abstract

Studies on fungal cellulase production via solid-state fermentation revealed that maximum enzyme production a mixture of wheat bran + rice straw 1:1 at 55% moisture content was observed on 4th day of incubation. 0.4% yeast extract, pH 5.0 was found to be optimum for high enzyme production. Using water as solvent maximum enzyme recovery was gained. The optimal pH and temperature for enzyme activity were 4.6 - 5.0 and 50 - 55°C. The crude enzyme was stable at pH 4.0-7.0 and at 40 - 60°C with more than 50% of activity when compared with control. Cellülàse and xylanase synthesized in solid fermentation was little more thermostable than that from liquid culture.
