STUDIES ON OCCURRENCE, TRANSMISSION AND DIAGNOSIS OF CITRUS YELLOW MOSAIC VIRUS (CYMV)

Abstract

Investigations on citrus yellow mosaic virus (CYMV) with regard to survey, transmission, diagnosis were conducted at AICRP on Tropical Fruits (Citrus), Tirupati during 2000-2004. Survey work revealed that CYMV was high in Ananthapur (24.8%) followed by Prakasam district (16.7%). Planococcus citri, a mealybug vector is able to transmit the virus, whereas the aphids failed to transmit it. Serodiagnosis, DAC-ELISA was standardized using the produced polyclonal antiserum. Detection of CYMV using the DNA extracted by addition of sodium sulphite in extraction buffer gave excellent amplification in PCR (726 bp). The produced polyclonal antiserum and PCR detection method are being used for indexing of nucellar and budling trees of Sathgudi sweet orange trees in the research station and also being used to diagnose the field samples collected during field diagnosis visits.
