PURIFICATION AND BIOCHEMICAL CHARACTERIZATION OF LIPASES THERMOPHILIC PSEUDOMONAS AERUGINOSA BTS-2P. GUPTA, R.K. KAUSHAL, S.S. CHIMNI, R. GUPTA AND S.S. KANWARAbstract Lipases (LC, 3.1,1,3.) that are capable of catalyzing hydrolysis, esterification and tran.s,esterification reactions both in aqueou.s and organic phases are finding increasing application in a number of Industrie'). Two extra-cell u r lipasc.as pases A and B) thermophilic Pseudomonac aeruginosa BTS2 were purified to homogeneity by ammonium '..ilfate precipitation and successive chromatographies on DEAF, cellulose and hydrophobic (Phenyl ,,epharose) column. The lipase A and B were found to be homogenous electophoretically with I4r of ?,0 and 42 kDa, respectively. The lipase A and B constituted approzimately .56 and 44%, of the total lipase activity and both pro,;;essed optima at ph I 7.5 and temperature 55" C. both lipases were highly specific' towards longer C-length p-nitro phenyl esters; however, they differed in their reaction kinOics.
