DEVELOPMENT OF IMMATURE GROUNDNUT (ARACHIS HYPOGAEA L.) CV. M-13 EMBRYOS IN VITROS.K. THIND AND NISHA BATRA
The developing embryos of groundnut (Arachis hypogaea L.) cv. M-13 were isolated at two stages of development (3 and 6 weeks after flowering from field grown crop and were inoculated on MS medium pH 5.8). The excised immature embryos (3 WAF) developed normally on BM-0.5 mg NAA + 0.25 mgl- + 0.1 mgl- ABA and produced vigorous plantlets. A different pattern of growth was observed when embryos excised 6 weeks after flowering were cultured on MS medium, as vigorous seedling were produced on BM, addition of NAA and GA3 reduced growth and ABA completely inhibited it. A pre-requisite for transferring genes into plants is the availability of a suitable plant regeneration system. The cell in culture displays its inherent potential to embark upon regenerative pathway that mimics with a high degree of fidelity in developmental programme of the zygote (Carman 1990). The lack of efficient regeneration systems for many grain le- gumes has slowed down the prog.ress in their genetic modifirai-;,,,, TT, work material from incompatible crosses (Ozias and Akir 1989). The high frequency of development an normal growth of the plantlets in general suggesi the presence of 'the required' levels of necessar growth regulators in the embryo axes.
