ALKALINE PROTEASE PRODUCTION AND OPTIMIZATION IN ESTUARY ISOLATE OF ASPERGILLLIS SP.

Abstract

The filamentous fungi, Aspergillus sp. was isolated from estuary soil samples. Protease production was confirmed by plating them on Skim Milk Agar plates. Optimization of the fermentation medium for mximum alkaline protease production was carried out. The chemical parameters like glucose, peptone, yeast extract and salt solution were optimized. The maximum enzyme activity under optimum condition was 960PU/L. The optimum values for the maximum alkaline protease production were; glucose 6(g/L), peptone 7(g/L), yeast extract 7(g/L) and salt solution 50 (ml/L). This procedure limited the number of actual experiments performed while allowing for possible interaction between the four components. This smaller and less consuming experimental design could generally suffice for the optimization of many fermentation processes.
