ISOLATION AND MOLECULAR CHARACTERIZATION OF BRUCELLA MELITENSIS FROM DAIRY PRODUCTS OF GOATS AND SHEEP IN RAPARIN DISTRICT, KURDISTAN REGION, IRAQ

Abstract

Brucellosis is a zoonotic disease that is found in animals and humans all over the world which is caused by Brucellaspecies. In this study which was performed to isolation and identification Brucellamelitensis from dairy products samples collected from some herds in Raparin district. In this study Serological test such as RBPT and 2ME test was used. Cultivation of bacteria was performed on some selective and deferential media such as Brain heart infusion broth, Brucella broth, Brucella agar, Colambiaagar and farallel selective medium, Isolation by using some biochemical tests such: (Oxidase, catalase,urease, blood hemolysis, and CO2 requirement) were done. A Polymerase Chain Reaction (PCR) was used for identification of Brucellaspp through detection bscp31 and IS711 genes from isolated bacteria. 130 positive of Rose Bengal Test were obtained which was performed previously on 520 blood samples that were collected from 36 herds of goats and sheep in this area. Among 144 total dairy product samples 14 isolates of Brucellamelitensis were cultured. Ten isolates from raw milk, 4 isolates from cheese samples and 2 isolates from human blood. For supporting the identification process, a molecular technique as confirmatory method through detection bscp31 and IS711 genes were performed. The prevalence of Brucellosis in livestock animals(sheep and goats) and human in Raparin district is endemic at significant rates and should be considered as an important health problem in this area.
